(630a) Transferring CHO Cells from Monolayer to Suspension Culture By a New Design of Experiments - Superlative Box Design

A new design of experiments – Superlative Box Design, is adopted to optimize the adaption of an anchorage-dependent, serum-dependent Chinese Hamster Ovary cell line to serum-free suspension culture. This seven-level quadratic response experimental design encompasses the uniformity in the distribution of experiments amongst all the factors of a central composite design while lower the number of experiments required. Six factors are applied in this experimental design with 43 runs plus three more repeating center runs. The cell line adapted to a serum-free medium by serum reduction, chemically defined media ratio increase, and mixing provided by rotation to reach suspension stepwise. Response surface methodology is applied to find the optimum condition.
