The spectrin is widely expressed nucleated cell and located in both cytoplasm and nuclear membrane. We analyzed the influence of spectrin in cytoplasmic membrane via knockdown alpha II spectrin. The results show the focal adhesion and actin fibers become more oriented after konckdown alpha II spectrin. The alpha II spectrin-lacking cells under shear stress are more sensitive and shown spectial response to the shear force. Our results suggest that spectrin network is important in cell mechanotransduction, adhesion and migration
