Details of excitation contraction coupling in skeletal muscle action remain under scrutiny, in part because real time measurements of molecular scale action transients remain challenging. The relationship between the nano-scale molecular events and the macroscale action is, however, largely massively parallel, which makes it possible to model the link between the nano-scale and the macro-scale in a relatively simple way as a first approximation. Further, this can be combined with the excitation process simply for appropriate values of Damkohler number for calcium activation.

Another observable issue is the filament compliance which appears to occur in a significant way in the pre powerstroke portion of the protein complex cycle.

Combination of these featues enables the integrated modeling of the protein cycle which allows for scaleup from the nnano-scale molecular response to the macro-scale tissue response. This can be used to extract nano-scale parameters from macro-scale data. These are testable model results.
