We will present an honest, “warts and all” history of our experience in bringing flow processing to the development of early stage candidate APIs. To become accepted flow technologies must enable the development chemist to undertake transformations deemed “unbatchable” for reasons of safety or extreme processing conditions, and we will present several case histories of these. However, we have found that technical issues are only one component in acceptance of the technology-the formidable skills of the synthetic chemist and the justifiably risk-averse development environment are steep barriers, for example. Considerable impediments exist for late-stage candidates as well. We will discuss our strategies for overcoming these barriers.
