Ceramic monoliths are a natural choice of support for the IMAC technique, because their large surface/volume ratios, low pressure drop during flow, strong mechanical properties and simple methods used for preparation and handling of active chromatographic columns. In addition monolith columns are relatively inexpensive and can be easily regenerated.

In this work the possibility multiple adsorption-desorption cycles on the separation efficiency of ceramic monoliths covered with agarose type D-5 using IMAC is investigated, using a well-known protein.. An adsorption model has been developed, in order to describe the dynamics of adsorption of proteins. The model is used to validate the experimental results obtained at equilibrium concentration and predict equilibrium concentrations of future cycles. The theoretical/experimental model developed in this research will allow determination of kinetic parameters and provide a tool for scaling up separation processes in industrial IMAC for a safe and effective use of ceramic monoliths as active media supports.
