(594r) Cell Culture Process Development for pH Sensitive CHO Culture

Initial 2L bioreactor studies for a high demand CHO product resulted in low titers and high lactate accumulation compared to previous shake flask screening studies. Trouble-shooting studies were performed to test differences between the bioreactor studies and the shake flask studies.  Mimicking the shake flask uncontrolled pH profile in both the inoculum train and production culture was found to increase titer and reduce residual lactate concentration. Optimization of pH conditions in all cell culture stages improved titers and metabolic behavior in bioreactors and resulted in more comparable performance to initial cell line screens.
