(632a) Targeted Delivery of Therapeutic Antioxidant Enzymes and Enzyme Mimetics

Oxidative insult by reactive oxygen species (ROS) is involved in development of a variety of conditions, such as secondary neuronal injury, arthritis, asthma, COPD, and many others. Natural antioxidant enzymes such as superoxide dismutase (SOD) are extremely efficient biocompatible ROS scavengers and as such could be of interest as therapeutic agents for these conditions. However, in most cases use of exogenous enzymes does not improve outcomes because of quick clearance in vivo and inability to achieve therapeutic concentrations at the site of injury. We developed tested a number of antibody-coated nanoparticulate vectors for targeted delivery  of SOD. In this talk we will discuss preparation and characterization of anti-NR1 antibody-coated SOD-nanoparticle conjugates for targeted delivery to central nervous system and report results of their testing as a therapy in a mouse stroke model.  We will also  discuss SOD- and antibody-coated nanoparticles for targeted delivery to respiratory tract, as well as their performance in mouse models of tobacco smoking and COPD.
