Andrographis paniculata or locally called hempedu bumi is a member of the plant family Acanthaceae. Andrographolide, a simple diterpene lactone, had been identified as the primary active chemical constituent in Andrographis paniculata extract that is responsible for the therapeutics of the plant. In this study, Response Surface Methodology (RSM) along with Box–Behnken design (BBD) was applied to optimize the ultrasonic-assisted extraction of andrographilde by investigating the combined influence of three independent variables which are extraction time (X1), duty cycle (X2) and ultrasonic amplitude (X3) on the andrographolide yield. The statistical analysis of variance indicated a high goodness of model fit (P <0.002; R20.9361). The suggested optimal conditions from the model for maximum andrographolide extraction were found to be extracted at 1.03 minutes, 21% duty cycle and 33A. The maximum andrographolide percentage experimentally was found to be very close to its predicted value (47.49 w/w%). Hence, the RSM method was successfully applied to optimize andrographolide ultrasonic-assisted extraction system.
