There is intense interest in using monoclonal antibodies as therapeutics because of their unique properties and activities. Nevertheless, there are many challenges in developing therapeutic antibodies related to their variable and difficult-to-predict propensities to self-associate, which can lead to a range of undesirable solution properties such as low solubility and abnormally high viscosity. I will discuss our recent progress in using self-interaction and phase behavior measurements of antibodies to understand and optimize their sequence-dependent thermodynamic properties.
