The high performance separation of membrane proteins has not yet been attained in despite of the significance, and been demanded. For that, we developed a novel separation method in a nano-lipid membrane, which utilizes the generated function of nano-lipid membranes.The nano-lipid membranes were constructed on the glass slide by self-assembly of lipid molecules. At first we investigated the factors affecting the separation in the nano-lipid membrane. In the nano-lipid membrane, membrane proteins kept its original structures as like in the cell membranes. In addition, the diffusion of membrane proteins was suppressed compared that in free solution. These specific properties of the nano-lipid membrane enable non-denaturing separation and high separation efficiency. Furthermore, we clarified that the separation efficiency of membrane proteins could be easily controlled by varying the composition and phase state of lipid membrane. Finally, we attempted the electrophoretic separation of membrane proteins and easily attained it in the nano-lipid membrane. Thus, we could successfully demonstrate the possibility of high performance separation in nano-lipid membranes and developed a new field of nano-separation sciences.
