In hybridoma cell cultures, amino acid and glucose consumption, cell growth and antibody production are key considerations during upstream process optimization, particularly media optimization. Gaining knowledge on their interrelations and the relevant cellular physiology could provide good insight for higher and more consistent antibody production. The amino acid consumption pattern was analyzed in hybridoma cell cultures producing monoclonal antibody (mAb). The amino acid concentrations, product concentrations, and cell growth profiles were measured in three different cell cultures subject to an amino acid supplementation strategy. In this research, we examine the amino acid metabolism and its impact on cell growth and mAb production during the different type of cell culture processes. We found that a group of key amino acids and metabolic pathways, the highest mass fraction of the product, and these could become limiting for product expression. In this study, metabolic pathway analysis was applied to show that certain metabolic pathways are significantly different between the culture process and media composition. Metabolic pathway analysis was helped to better evaluate the impact of amino acid supplementation on the most relevant pathways in the culture process.
