Immobilization is a technique used to prevent the contamination of active enzymes, increase substrate mobility and to prevent cell washout. Immobilization has shown its success in Submerged Fermentation (SmF) because of the liquid media that is used but the shear effects of solid substrates prevents its usage in Solid State Fermentation (SSF). In a first of its kind study, an extract of the solid substrate was obtained using buffer solution and this extract is treated as liquid media and SmF process is run using calcium alginate beads as the immobilized material. L-Asparaginase is the most common enzyme used in the preparation of medicinal drugs used in the treatment of Acute Lymphoblastic Leukaemia (ALL). It can also be put to use in the food industry to reduce the carcinogenic effects of acrylamide that is present in deep fried foods. SSF has been always observed as an alternative to the SmF because of its higher benefits which include economic and environmental factors. Optimization of the parameters will help in further improvement of the activity and different techniques can be tested to bring out the best result. Different solid substrate extracts were tested and the best out of it was selected as wheat bran extract, the activity of wheat bran substrate is expected to be around 11 U/ml with a specific activity of 10.5 U/mg in case of the extract media as opposed to 4-5 U/gds in the solid media. The diffusion parameters that are involved in the study include the intra-particle and external diffusion in the immobilized beads, these shall also be studied and an understanding of the factors will be reported.
