G-protein coupled receptors (GPCRs) are a highly desirable target for drug development due to their broad-range of signaling effects and roles in many diseases such as cancer, infection, and autoimmune disorders. Approximately 34% of all FDA approved drugs target GPCRs, but there are currently no antibody-based GPCR therapeutics in clinical use. Antibody discovery against GPCRs is a challenging task due to the difficulty in recombinantly expressing the glycosylated seven-transmembrane domain proteins in their native state. Here we couple the exciting new virion display platform presenting GPCRs in their functional conformations with the yeast surface display directed evolution platform for high-throughput anti-GPCR antibody discovery.
