RNA is one of the key molecules in cell. It is a messenger, a carrier, an enzyme and unknown role. Some diseases are resulted from RNA changes, such as editing, truncation or alternative splicing. The detection of RNA is more difficult from their short half life.
For monitoring the RNA, adaptors with assigned nucleotides were adhered in both ends before reverse transcribed. The the target gene was amplified with gene-specific primer as well as the primer complentary to the adaptor with cDNAs. And then the amplicons were cloned and sequenced. The modification of RNA was clarified. The technique was aplied in the cells treated with siRNA, aninal model and patients. The trucation site of RNA from samples were identifed.
