(481h) Chemical Processes Optimization By Hydrodynamic Simulations – Implementation in the Solid Phase Peptide Synthesis

In this presentation, we will describe the concept of Scale Down – Scale Up (SDSU) for Batch and Semi Batch operations in stirred reactor systems. The concept is based on hydrodynamic studies of the mixing procedure obtained from experiments in small scale mixed rectors and process simulation by VisiMix software. We describe how the hydrodynamic simulations on small and large scale reactions can aid in process optimization and scale up. The use of the SDSU concept and methods described in this article, will offer the user the possibility to achieve the best results during production stage, saving time and currency, and at the same time increasing the knowledge of the performed process. Several examples in the article demonstrate the benefits of the proposed method.

The brittle nature of the polymer beads used in SPPS dictates mild mixing techniques with low mass transfer. We demonstrate that using vigorous overhead mechanical stirring with superior mass transfer properties kept the beads intact and significantly accelerates reaction kinetics and efficiency.
