Systems metabolic engineering using in silico tools has facilitated mass production of wide range of valuable chemicals. Elementary Mode (EM) analysis, one of the useful in silico tool, has the advantage of simple implementation, however, there are difficulties with analysis and application of thousands of EMs for metabolic engineering. Here, we designed method by selection of optimal EMs and subsequently combining of clustering method, thereby easily provides valuable information for metabolic engineering. EM analysis with clustering method applied to analyze optimal metabolic network for the production of succinic acid in Mannheimia succiniciproducens, and simulation results were validated with previous experiment results. Additionally, the yield of succinic acid production in M. succiniciproducens successfully improved through experiments from simulation results.
